Transfection in Flow

Fast SnapFect cell transfection with superior viability and efficiency

New way to transfect cells with RNA, DNA, CRISPR

Rapid SnapFect Transfection in Flow. (A) The bio-orthogonal mediated nucleic acid transfection strategy is fast and can be performed in a microfluidic format. The mixing of the keto tailored cells with the nucleic acid lipoplex for transfection occurs in less than 1 minute. The cells are then visualized after 24 hours and show high viability and efficiency of transfection. The entire experiment was replicated over 100 times, run independently and the images compared to controls where one or both pairs of the click chemistry groups are missing from the surface of the cell or the nucleic acid complex. (B) Red fluorescent protein expressing fibroblasts that are transfected with GFP in the microfluidic format. The cells turn yellow and green after GFP transfection via the fast microfluidic method (C). (D) Nuclei stained 3T3 fibroblasts are flowed through the microfluidic channel and mixed with GFP nucleic acid lipoplex (1 min). (E) The fibroblasts are visualized after 24 hours and are efficiently transfected and show green fluorescence with high viability. Each microfluidic transfection experiment was replicated over 100 times and analyzed by fluorescence imaging and protein gel analysis and compared to controls where one or both pairs of the click chemistry pair are missing from the surface of the cell or in the nucleic acid complex.
